Which regulation requires it

Regulatory Reform (Fire Safety) Order 2005. In England and Wales the Order places duties on the "responsible person" for non-domestic premises, including a duty under Article 14 to keep emergency routes and exits clear and indicated. Scotland is covered by the Fire (Scotland) Act 2005 and the Fire Safety (Scotland) Regulations 2006, and Northern Ireland by the Fire and Rescue Services (Northern Ireland) Order 2006. Escape route signage is normally specified to BS 5499-4 alongside the ISO 7010 graphical symbols.
